Il a détenu au cours de sa carrière professionnelle le titre de champion du monde des poids lourds de 1926 à 1928, ainsi que le titre de champion des poids mi-lourds des Etats-Unis à deux reprises entre 1922 et 1923.Boxeur hautement technique, son ascension après la Première Guerre Mondiale lui a permis de s'opposer aux boxeurs les plus célèbres de sa génération. Il est notamment connu pour ses oppositions face à Harry Greb. Celles-ci se sont déroulées au cours de cinq combats en poids mi-lourds ; Tunney en a remporté trois, fait un match nul et perdu une fois. Leur seconde rencontre, remportée par Tunney, a toutefois fait l'objet de controverse, de nombreux journalistes au bord du ring ont en effet vu Greb gagnant,.
